Mechanisms of Protein Import into Chloroplasts

研究室簡介:

大多數的葉綠體蛋白質是在細胞質中合成,然後送入葉綠體。這些蛋白質能否被正確地送入葉綠體乃是決定葉綠體能否正常運作的關鍵之一。由細胞質送往葉綠體的蛋白質大致可分為兩大類(如左圖),第一類為進入葉綠體內部的蛋白,第二類為葉綠體外膜蛋白。要進入內部的蛋白,在細胞質合成時,氨基端皆多了一段導引訊息(transit peptide)。他們進入葉綠體的過程需要ATP 及一組運輸機組(translocon)的幫助。而要送往葉綠體外膜的蛋白,在細胞質合成時,大多在氨基端並沒有多一段導引訊息,且他們插入外膜的過程中,早期研究認為不需要ATP 或其它蛋白的協助。

 

我們實驗室已從兩個葉綠體的外膜蛋白找出它們內含的導引訊息,並發現外膜蛋白和內部蛋白的運輸途徑交會於外膜通道Toc75 上。此外我們也以遺傳學的方法研究蛋白質輸入葉綠體內部的機制,由對各種變株的研究,我們發現某一葉綠體特有脂肪對蛋自質輸入葉綠體的重要性,以及運輸機組乃受植物發育及組織細胞調控。我們並分離到一調控運輸機組蛋白基因轉錄的因子。再藉由研究各個運輸機組成員之間的組合與交互作用,我們正逐步呈現蛋白質進入葉綠體的連續步驟 (如左圖)。
